Real-Time PCR Work flow












• Choose from singleplex, multiplex, low-throughput and high-throughput
Chemistry Selection qPCR depending on the type of analysis and decide according to purpose
(DNA binding Dyes whether to use DNA binding dyes, uorescent primer or probe type de-

• In case of low-throughput singleplex qPCR, use DNA-binding dye such as
SYBR® Green I or EvaGreen™ Dye and in case of high-throughput qPCR, use

What is Passive Reference



Dye? (e.g. ROX,Fluorescein dye etc.)







Passive reference dye is used as a reference point to correct the uorescence signal in real-time PCR or to calculate the uctuation
of the uorescence signal. Calibration means modifying to minimize the di erence between the total volume or the equipment
scanner during the reaction. In general, most real-time PCR equipment uses ROX dye as a passive reference dye. This is because
ROX dye does not have any e ect on real-time PCR reaction and emits a uorescence signal that is clearly distinguished from probe
uorescent dye (reporter or quencher). As with some equipment from Bio-Rad, there are some that use uorescein dye and there
are systems that do not require a passive dye such as the Rotor-gene. It is recommended to select the passive reference dye to be
used after considering the advantages and disadvantages of each device or method.

pli cation and users can use their own intercalating dyes by themselves. Furthermore, EvaGreen™ or SYBR® Green I can be supplied
with our kits as an option or mixture types for convenient use and included BioFACT™ H-Star Taq DNA Polymerase helps accurate and
reliable result to enable target-speci c ampli cation due to its Hot Start characteristic.
BioFACT™ 2X Real-Time PCR Master Mix and Pre-Mix contains H-Star Taq DNA polymerase, dNTPs, optimized reaction bu er, passive

Outstanding Intercalating Dye 100% Made in Korea






SFCgreen® I is a green uorescent dye with excitation and emission wavelengths very similar to those of SYBR® green I or Eva-
Green™. The DNA binding mechanism is a bis-intercalator that binds to the DNA minor groove and emits light.
SFCgreen® I has high DNA binding strength and shows a higher sensitivity compared to SYBR® green I or EvaGreen™. Because
PCR inhibition is low, PCR ampli cation e ciency is high even at high concentrations, and it is stable under light, heat, and
chemicals, resulting in a longer uorescence half-life compared to other uorescent dyes.

BioFACT™ 2X Real-time PCR Kit using SFCgreen® I contains H-Star Taq DNA polymerase with Hot Start PCR function, so that only target
DNA is speci cally ampli ed. BioFACT™ 2X Real-time PCR Kit is a pre-mixed form of H-Star Taq DNA Polymerase, dNTP, Reaction bu er
and SFCgreen® I, so that it can be conveniently used by simply adding the primer and the template to perform qPCR rection.
The use of passive reference dye di ers depending on the model, so we have produced a variety of products suitable for many models
to increase user convenience. Also, for pre-mixed products, it is available in three types: 0.1㎖ 8-strip Frosted Tube, White Tube, and Clear
Tube, so it can be used in various models.

BioFACT™ 2X Real-time PCR Kit using SFCgreen® I contains A-Star Taq DNA polymerase with hot start PCR function, so that only
target DNA is speci cally ampli ed. BioFACT™ A-Star 2X Real-time PCR Kit is a pre-mixed type of A-Star Taq DNA Polymerase, dNTP,
Reaction bu er and SFCgreen® I, so that it can be conveniently used by simply adding the primer and the template to perform
qPCR reaction. The use of passive reference dye di ers depending on the model, so we have produced a variety of products suita-
ble for many models to increase user convenience. Also, for pre-mixed products, it is available in three types: 0.1㎖ 8-strip Frosted
Tube, White Tube, and Clear Tube, so it can be used in various models.

20X EvaGreen™



Intercalating Dye with High Sensitivity and Stability







20X EvaGreen™ is the next generating DNA binding dye that can be used in Real-Time PCR. EvaGreen™ is compatible with HRM
analysis as it has less PCR interference, such as slowing down the primer dimer formation and extension reaction, as well as high
sensitivity to double-stranded DNA, and can produce more accurate results. It has little PCR inhibition even at high concentrations
and is compatible with most qPCR instruments in a wavelength similar to SYBR® Green I. It is non-mutagenic, noncytotoxic, and very
stable under various PCR conditions.

F-Star Taq DNA Polymerase


Optimal Taq DNA Polymerase for Less Than 3kb Speci c PCR






BioFACT™ F-Star Taq DNA Polymerase is designed to be highly speci c to the target gene through chemical-mediated Hot Start
system. It has similar features with BioFACT™ H-Star Taq DNA Polymerase and especially it is suitable to amplify DNA fragments larger
than 1kb. In case of chemical-mediated Hot Start PCR enzymes, its ampli cation e ciency and productivity may be lower than gen-
eral Taq DNA Polymerase. In this case, it can be supplemented by increasing the cycle.

Highly Speci c Ampli cation of Less Than 1 kb Size Target DNA






BioFACT™ H-Star Taq DNA Polymerase is a chemical-mediated Hot Start Taq DNA polymerase which leads to complete inactivation of
the polymerase until the initial heat activation step. Through this feature, it provides high PCR speci city, avoiding extension of non
speci c annealed primers or primer-dimers and it is highly suitable for e cient ampli cation of di cult templates caused by high GC
content or complex structure. BioFACT™ H-Star Taq DNA Polymerase is optimized to amplify less than 1 kb size target DNA and it can
be supplemented by increasing the cycle as ampli cation e ciency and productivity of chemical-mediated Hot Start PCR enzymes
may be lower than general Taq DNA Polymerase.

A-Star Taq DNA Polymerase


Antibody-Mediated Hot Start PCR Enzyme






BioFACT™ A-Star Taq DNA Polymerase, isolated from Thermus aquaticus and expressed in E.coli through a simple and modi ed puri ca-
tion procedure, is Hot Start PCR enzyme which is combined with anti-Taq monoclonal antibody that inhibits polymerase activity until it
is heat denatured. It has shorter pre-denaturation time than chemical-modi ed Hot Start PCR enzyme and it also prevents non speci c
ampli cation and primer-dimer formation. BioFACT™ A-Star Taq DNA Polymerase is highly suitable for ampli cation reactions involving
low-copy targets, complex genomic DNA or cDNA templates. Providing optional 5X Band Helper™ helps ampli cation of di cult PCR

Lamp Pfu DNA Polymerase


Blunt-End Fragment Ampli cation of Less than 20 kb with High Accuracy






BioFACT™ Lamp Pfu DNA Polymerase is an upgraded version of existing Pfu DNA polymerase using mutagenesis to express the Pyro-
coccus furiosus DNA polymerase gene in E.coli. While retaining the features of BioFACT™ Pfu DNA Polymerase, the extension time is
signi cantly reduced to 30 seconds per 1kb and the ampli cation rate is much better than that of Pfu DNA Polymerase.
In addition, it enables the ampli cation of long PCR fragments of 20 kb (based on human gDNA). This enzyme also allows blunt
cloning without an additional A-tailing process using the All in One™ PCR Cloning Kit.
