kaedah konvensional. Ini kerana, kaedah ini dapat menjamin pembiakan yang bebas penyakit,
benih yang dihasilkan lebih terjamin daripada segi ketulenan varieti, pokok tumbuh seragam, lebih
cepat dan boleh dilakukan secara skala besar. Kaedah kultur tisu melibatkan penggunaan
kemudahan makmal yang menyediakan ruang kerja yang steril, aseptik atau bebas kuman.
Pembiakan pisang melalui kaedah kultur tisu bermula daripada pemilihan dan penggunaan sulur
daripada pokok induk yang baik. Pokok induk yang baik adalah yang bebas daripada penyakit,
berprestasi baik serta berupaya mengeluarkan hasil yang tinggi. Sulur yang digunakan ialah sulur
pedang kerana ianya mempunyai keupayaan hidup dan membiak yang tinggi selepas dikultur
secara aseptik. Keratan sulur pedang akan melalui proses pensterilan permukaan menggunakan
agen-agen pensterilan bagi menyingkirkan mikroorganisma seperti kulat, bakteria serta virus dan
seterusnya meminimakan risiko kontaminasi. Eksplan tisu meristem daripada keratan sulur yang
telah steril akan dikulturkan secara aseptik bagi tujuan pencambahan dan multiplikasi secara
berkala.
Abstract
Propagation of banana crops using tissue culture method is more effective than conventional
method. This tissue culture method can guarantee disease-free reproduction, seedlings produced
are true to type, grow uniformly, faster and can be done on a large-scale. Tissue culture methods
involve the use of laboratory facilities that provide a sterile, aseptic or germ-free workspace.
Propagation of bananas through tissue culture method starts from the selection and use of
suckers from good quality parent clones. A good parent clone is one that is free from disease,
performs well and is able to produce high yields. The suckers used are sword suckers because
they have a high ability to survive and reproduce after aseptically cultured. Sword sucker cuttings
will go through a surface sterilization process using sterilization agents to get rid of
microorganisms such as fungi, bacteria and viruses and thus minimize the risk of contamination.
Meristem tissue explants from sterile sucker sections will be cultured aseptically for continuous
proliferation and multiplication.

Abstrak
Dasar Pertanian Nasional (DPN 3) 1998-2010 dan Dasar Agro Makanan Nasional (NAP) 2011-
2020 adalah bukti usaha kerajaan Malaysia dalam mengubah dan meningkatkan kualiti makanan
negara. Dasar Agrofood Nasional 2011- 2020 digubal dengan fokus khusus untuk meningkatkan
sektor pengeluaran makanan, termasuk beras. Walaupun Malaysia masih bergantung pada
beras import untuk memenuhi permintaan pengguna, industri beras selalu menjadi keutamaan
nasional berdasarkan kepentingan strateginya sebagai komoditi makanan tanaman. Walaupun
begitu, industri padi di Malaysia dikekang oleh beberapa cabaran seperti perubahan iklim global,
kekurangan variasi baru, benih yang diperakui tidak mencukupi, munculnya penyakit utama
seperti Hawar Bulir Bakteria (BPB), karah daun, musim kemarau, banjir kilat, penurunan kawasan
penanaman, kehilangan minat petani kerana kos pengeluaran padi yang tinggi dan banyak lagi.
Oleh itu, NAP telah menekankan bahawa pengeluaran beras tempatan harus ditingkatkan untuk
memastikan permintaan negara pada masa hadapan. Di samping itu, dari 2017 hingga 2020,
beberapa ujian penentusahan setempat (LVT) juga dilakukan dari bahagian utara hingga
bahagian selatan Semenanjung Malaysia. Data yang diperoleh dari Sekinchan, Selangor
menunjukkan bahawa NMR 152 secara konsisten menghasilkan antara 7 - 10 t/ha di kawasan
jelapang padi berbanding dengan 6 t/ha yang dihasilkan oleh varieti lain di kawasan penanaman
yang sama. Pengujian di lapangan juga menunjukkan bahawa kos pengeluaran dikurangkan
sebanyak 10%, disebabkan oleh penurunan penggunaan baja dan racun perosak. Pada masa
yang sama, hasil dapat ditingkatkan antara 40 - 67% bergantung pada kawasan penanaman. Di
samping itu, dua titisan mutan berjaya diberikan Sijil Pendaftaran Varieti Tumbuhan Baru dan
Pemberian Hak Pembiak oleh Jabatan Pertanian Malaysia pada Februari 2020 dengan nombor
pendaftaran; PBR0156 (untuk NMR152) dan PBR 0159 (untuk NMR151).
Selain meningkatkan hasil dan pendapatan petani, titisan mutan yang berasal daripada projek
FNCA juga sangat mempengaruhi status sosio-ekonomi petani kerana padi mutan dapat
disesuaikan dengan keadaan perubahan iklim global ketika ini. Selanjutnya, melalui MoU dengan
syarikat-syarikat, sekitar 4.200 metrik tan benih telah dihasilkan pada tahun 2020 untuk
penyediaan 25.833 hektar kawasan penanaman di Semenanjung Malaysia. Pada 10 Ogos 2020,
Kementerian Pertanian dan Industri Makanan (MAFI) telah mengesahkan NMR152 sebagai
varieti padi baru nasional setelah NMR152 menjalani pembentangan teknikal dengan
Jawatankuasa teknikal Bantuan Kerajaan Kepada Industri Padi dan Beras (BKKIPB) BIL.2 /
2020). Pada asasnya, projek ini telah membahas agenda dan dasar nasional dalam penghasilan
varieti padi baru untuk meningkatkan kesejahteraan dan taraf hidup petani.
Abstract
National Agriculture Policy (DPN 3) 1998-2010 and the National Agrofood Policy (NAP) 2011-
2020 are the evidence of Malaysian government's efforts in transforming as well as improving the
country's food quality. The National Agrofood Policy 2011-2020 was formulated with a special
focus on improving the food production sector, including rice. Even though Malaysia is still
depending on imported rice to fulfil consumer's demand, rice industry has always been a national
priority based on its strategic importance as a staple food commodity. Nevertheless, the rice
industry in Malaysia is hampered by several challenges such as global climate change, lacking of
new variety, insufficient certified seed, emerging of major diseases such as Bacterial Panicle
Blight (BPB), Leaf Blast, drought season, flash flooding, decreasing of planting area, lost interest
of the farmers due to high cost of rice production and many others.
Thus, the NAP had highlighted that local rice production should be increased to ensure the
country's demand in the future. In addition, from 2017 until 2020, several local verification trails
(LVT) were also conducted from the Northern part until the Southern part of Peninsular Malaysia.
The data obtained from Sekinchan, Selangor revealed that NMR 152 consistently produced
between 7-10 t/ha in granary area as compared to 6 t/ha produced by other varieties within the
same planting areas. Field trials also showed that the production cost was reduced by 10%,
mainly due to the reduction in fertilizer and pesticide usage. At the same time, the yield could be
increased between 40 - 67% depending on the planting areas. In addition, two mutant lines were
successfully granted with Certificate of Registration of New Plant Variety and Grant of Breeder's
Right by Department of Agriculture Malaysia in Feb 2020 with registration number; PBR0156 (for
NMR152) and PBR 0159 (for NMR151).
Apart from increasing the yield and income of the farmers, the mutant lines derived from FNCA
project had also greatly impacted the socio-economic status of the farmers as the mutant rice are
adaptable to current global climate change conditions. Furthermore, through the MoU with the
companies, around 4,200 metric tons of seeds have been produced in 2020 for the supply of
25,833ha planting areas in Peninsular Malaysia. On 10 August 2020, Ministry of Agriculture and
Food Industries (MAFI) will certify NMR152 as national new rice variety after NMR152 undergone
the technical defense with the technical committee (JawatanKuasa Teknikal Bantuan Kerajaan
Kepada Industri Padi dan Beras (BKKIPB) BIL.2/2020). Basically, the project has addressed the
national agenda and policy in generating new rice varieties and thus, increase the well-being and
livelihood of the farmers.

Abstrak
Isotop stabil semakin banyak digunakan sebagai penyurih dalam kajian persekitaran. Salah satu
aplikasi adalah menggunakan nisbah isotop untuk menentukan secara kuantitatif perkadaran
sumbangan beberapa sumber terhadap campuran, seperti perkadaran pelbagai sumber
pencemaran dalam aliran air. Dalam beberapa dekad kebelakangan ini, analisis dual isotop nitrat
(15N dan18O) dan sulfat (34S dan18O) telah banyak digunakan untuk mengesan sumber
pencemaran. Aplikasi umum adalah menggunakan komposisi isotop campuran untuk
menentukan perkadaran pelbagai sumber dalam campuran, menggunakan model pencampuran
matematik. Bagi melakukan pensampelan air di lapangan, penyelidik perlu merancang dan
merancang aktiviti dengan baik. Lawatan tapak dan pemetaan lokasi perlu dilakukan terlebih
dahulu untuk mengenal pasti pernyataan masalah di kawasan kajian. Titik persampelan perlu
diatur agar sesuai dengan model yang akan digunakan. Peralatan dan alatan persampelan mesti
disediakan dengan secukupnya agar kerja persampelan dapat dijalankan dengan lancar. Kertas
kerja ini akan menghuraikan prosedur operasi standard (SOP) Pensampelan air untuk analisis
isotop stabil di lapangan.
Abstract
Stable isotopes are increasingly being used as tracers in environmental studies. One application
is to use isotopic ratios to quantitatively determine the proportional contribution of several sources
to a mixture, such as the proportion of various pollution sources in a water stream. In recent
decades, the dual isotope analysis (15N and18O) of nitrate and (34S and18O) of sulfate has been
widely applied to track contamination sources. A common application is to use the isotopic
composition of a mixture to determine the proportions of various sources in the mixture, using
mathematical mixing models. To conduct water sampling in the field, researchers need to design
and plan well the activity. Site visits and mapping need to be done first to identify the problem
statements of the study area. Sampling points need to be set so that they are in line with the
model to be used. Sampling equipment and tools must be provided adequately so that the
sampling work is carried out smoothly. This paper will elaborate standard operating procedure
(SOP) of water sampling for stable Isotope analysis in the field.

CRUCIAL STAGE IN INITIATING DIFFERENT TYPES OF EDIBLE MUSHROOMS

Abstrak
Cendawan tiram kelabu (Pleurotus pulmonarius), cendawan jeli hitam (Auricularia auricula) dan
cendawan jerami padi (Volvariella volvacea) adalah spesies cendawan yang boleh dimakan yang
paling biasa dinikmati oleh rakyat Malaysia kerana sifat rasa yang baik dan kandungan nutrien
yang tinggi. Induk utama atau biasanya dikenali sebagai ibu tisu didik adalah elemen penting
dalam menghasilkan anak benih cendawan yang baik dan berkualiti. Teknik tisu kultur secara in
vitro dalam memulakan pemencilan tisu dalam menyediakan ibu tisu adalah langkah yang paling
penting. Pertama, pemeriksaan dan pemilihan jasad buah berkualiti yang digunakan untuk
memulakan pemencilan tisu mestilah segar, sihat dan bebas daripada sebarang tanda-tanda
perosak dan penyakit. Kedua, tempat melakukan aktiviti tisu didik mestilah tertutup dalam
lingkungan, bersih dan selesa seperti di dalam kebuk aliran udara atau ruang bersih. Persiapan
untuk ruangan bersih perlu dilakukan 30 minit sebelum kerja-kerja pemencilan tisu dilakukan.
Ketiga, alat untuk memotong tisu mestilah kering, steril dan biasanya di autoklaf pada suhu
121°C, 15 psi selama 15 minit. Selain itu, penggunaan media dalam menumbuhkan miselium
harus disiapkan dalam keadaan bebas daripada bakteria dan/atau fungus. Individu yang
melakukan aktiviti itu perlulah bersih, dilengkapi dengan baju makmal dan sihat. Proses
mengeksekusi tisu harus dilakukan dengan berhati-hati dan lembut. Semua langkah yang
disebutkan adalah mustahak untuk mendapatkan sumber asas tisu didik yang baik dan bebas
dari pencemaran. Kertas kerja ini membincangkan secara mendalam langkah dan prosedur yang
diambil dalam memulakan pelbagai jenis cendawan serta kadar pertumbuhan dan prestasi
pertumbuhan miselium untuk setiap spesies cendawan.
Abstract
Grey oyster mushroom (Pleurotus pulmonarius), black jelly mushroom (Auricularia auricula) and
paddy straw mushroom (Volvariella volvacea) are the most common andtypicaledible mushrooms
species enjoyed by Malaysian owing to their good flavors properties and high nutrients content.
Master culture or commonly known as mother culture is a crucial element in producing a good
mushroom seedling. In vitro technique of initiating tissues in preparing the mother culture is the
most crucial step. Firstly, screening and selection for quality fruiting body used to initiate tissue
must be fresh, healthy and free from any spots of pest and diseases. Secondly, place in doing
the activity must be closed environment, clean and comfortable such as in laminar air flow or
clean chamber. Preparation for the clean area is conducted 30 minutes prior to tissue excises.
Thirdly, tools in dissecting the tissues must be dry, sterile and normally autoclaved at 121°C, 15
psi for 15 minutes. Besides, media used in growing the mycelium should be freshly prepared and
free from any spot from bacteria and/or fungus. Fourthly, personnel carrying out the activities
must be clean, equipped with laboratory coat and healthy. Fifthly, the process in excising the
tissues should be conducted carefully and gently. All steps mentioned are important in getting a
good material free from contamination. This paper discusses in depth the steps and procedures
taken in initiating different types of mushrooms as well as their growth rate and growth
performance for each mushroom species.
