Solution-Phase Affinity & Kinetics

For over 25 years KinExA Technology has been used to detect & measure
biological interactions in the most physiological relevant conditions with
precision. Industry leading accuracy and sensitivity into the femtomolar range
make KinExA an integral part of the lead identification and optimization process.

KinExA 4000

The KinExA 4000 requires low sample volumes and can accomodate up to 4 distinct
solid phase preparations with up to 270 samples that will support extended periods

of autonomous operation. Intuitive software combined with accurate and diverse
measurement capabailites make the 4000 a reliable instrument for your lab.

Purchase includes a 1 year warranty, 10 year LED warranty, 2 licenses for KinExA
Pro software, installation, training, and unlimited GxP support. Ditch the chip and

enjoy low cost consumables compared to other platforms.

Trusted Accuracy | Proven Reliability | Unrivaled Sensitivity

KinExA Pro Software

KinExA Pro Software is a robust tool for analyzing biomolecular interactions from data
generated with KinExA instruments. With a single click, exact solutions to the binding
equations are used to fit a theoretical binding curve to experimental data. Along with
affinity and kinetics data, researchers will get useful information on copy number of

receptors on cell surfaces, activity regarding one of the binding partners, unknown
concentrations, and the amount of error between the theory curve and experimental data

(residual error). The N-curve analysis includes curves at varying concentrations that
helps to exploit discrepancies in the binding behavior.

95 % confidence intervals are generated
simultaneously with binding curves that
show how well parameters like equilibrium
dissocation constant or activity are defined.

“ To gain a more quantitative measurement of the antibody binding affinities to guide our epitope mapping
experiments, we turned to KinExA analysis, a method that measures equilibrium binding affinities of unmodified
molecules in the solution phase. ”

Wang X. et. al. 2017. Fine epitope mapping of two antibodies neutralizing the bordetella adeynlate cyclase toxin. Biochemistry. 56 (9). pp 1324-1336

“ Having a robust methodology for characterizing antibody interaction with a membrane protein target is essential for
providing guidance on dosing, potency, and thus expected efficacy...our study concluded that KinExA enables accurate
measurement of the KD for strong protein-protein interactions (sub-nanomolar values) compared to FACS. ”

Vaish A. et. al. 2020. Binding affinity determination of therapeutic antibodies to membrane targets:
KinExA using cellular membranes for anti-CD20 antibody. Analytical Biochemistry. vol 609

“ KinExA is particularly attractive to study protein lipid interactions because native untagged molecules binding
entirely in solution can be investigated. “

Fleming JK, et. al. (2016) A novel approach for measuring sphingosine-1-phosphate and lysophosphatidic acid binding to carrier proteins
using monoclonal antibodies and the kinetic exclusion assay. J Lipid Res 57(9):1737–1747. doi: 10.1194/jlr.D068866

“ The kinetics of binding and affinity of the 4 chimeric clones to human IL-21 were determined using a kinetic exclusion
assay (KinExA)...Among the four lead clones, 19E3 had the highest affinity to recombinant human IL-21 of 243 fM. ”

antigen immunization and humanization strategy. PLoS ONE. 14(1): e0211236. https://doi.org/10.1371/journal.pone.0211236

“ Although numerous laboratory techniques have been developed to study the intricate interactions between proteins
and DNA, each one of them has limited utility regarding applications. For better quantification of these important
interactions, our work focused on using the Kinetics Exclusion Assay (Kinexa) technology. ”

Leung, E. et al. 2018. Quantifying protein-DNA interactions by kinetic exclusion assay. Biophysical Journal, 114(3): 442a

Temperature Control for KinExA research

The TC1000 gives you the ability to incubate samples & run experiments at any
temperature between 4⁰C and 40⁰C. The wide temperature range and an

integrated computer with KinExA Pro software installed gives you the ability to
estimate thermodynamic parameters of binding.
