Luxturna, a new gene therapy, has been approved by the US Food and Drug Administration (FDA) to treat an inherited form of vision loss that can result in blindness. It is the first directly administered gene therapy approved in the US that targets a disease caused by mutations in a specific gene. The RPE65 gene provides instructions for making an enzyme that is essential for normal vision. Mutations in this gene lead to reduced or absent levels of RPE65 activity, blocking the visual cycle and resulting in impaired vision. Luxturna has been approved for the treatment of patients with confirmed biallelic RPE65 mutation-associated retinal dystrophy that leads to vision loss and may cause complete blindness in certain patients. This is a rare disease affecting around 1,000 to 2,000 patients in the US. Luxturna is the third gene therapy approval in the US in 2017, indicating a new frontier for treating diseases. OUTBREAK NEWS ON HUMAN INFECTION CAUSED BY AVIAN INFLUENZA Epidemiology Description of this case Researcher are form a report on this disease through their study by WHO (World Health Organization) and recently published on 6 April 2023 infection caused by avian influenza A(H5N1) in Chile. Avian influenza is a virus which is caused by a virus of the Orthomyxoviridae family and it is also known as “bird flu”. Avian influenza can cause severe disease in human , it is notifiable under International Health pRegulation (IHR 2005). The outbreak of avian influenza virus are responsible for a sharp increase of disease in poultry and other bird in the recent year. On 5th April, the Institute of Public Health of Chile which is the National Influenza Centre in Chile has reported the result of genomic sequencing in which they identified that avian influenza virus A(H5N1) phylogenetic clade. The genomic sequences had 99.9% idenfied with H5 hemagglutinin sequences from Chilean bird and complete neuraminidase has 100% identify with N1 sequence from Chilean bird. The zoonotic influenza infection may be asymptomatic at primary stage in human and show mild respiratory infection or fever and cough to rapid continuation in severe pneumonia, acute respiratory distress syndrome, shock, and at the last stage it causes death that are depending upon the various factor related to virus and host. Sometime, gastrointestinal or neurological symptoms were reported in the previous study. In human being the avian influenza virus were spread through live or dead poultry or contaminated environment. During 2022 and 2023, in the region of America increasing the number of outbreak huge pathogenic avian influenza A have been reported mostly from the area of poultry farm, wild birds and wild mammals. Since the first confirmation of avian influenza(H5N1) in the region in 2014 three human infection caused by avian influenza (H5)have been reported i.e., first in the United State of America, reported in April 2022; the second case reported in Ecuador in January 2023; and the third is reported in 2023 in the America. As per the report, on 13 March 2023, mild symptoms of avian influenza were shown in a patient like cough, sore throat and hoarseness then on 21 March, due to worsening symptoms he sought care at a local hospital. On 22 March, the that patient developed dyspnea and was admitted to a Regional Hospital at Antofagasta then his nasopharyngeal swab sample was collected as a part of routine severe acute respiratory Pharma Bulletin ISSUE NO. On 23 March, patient were admitted to the intensive care unit and treatment with antiviral and antibiotics was initiated. Then that patient remain in respiratory isolation under multidisciplinary management with mechanical ventilation due to pneumonia. Standard infection control precaution were kept since the detection of the case. On 5th April, the result of genomics sequencing were completed by NIC in Chile, which identified avian influenza virus A(H5N1) phylogenetic clade 2.3.4b. The genomic sequence had 99.9% identified with H5 hemagglutinin sequences from Chilean birds and complete neuraminidase (NA) had 100% identity with N1 sequences from Chilean birds. Another a total nine cases were reported in healthcare worker and then they tested negative for avian influenza and on 4th April ended the monitoring period. On the 5th April there were one person had shown respiratory symptoms , therefore a nasopharyngeal swab was taken , and tested negative and the monitoring period for this contact was extended for seven days and then this case ended on 11 April. Recent Discovery in Delivery of PEGylated liposomal doxorubicin by bispecific antibodies improves treatment in high-risk childhood leukemia This is an auspicious invention to improve the treatment for high-risk childhood leukemia. This article, published in science translational medicine which mainly focused on improving biodistribution and tolerability of chemotherapy. Drug encapsulation with liposomal carrier has shown clinical success. In previous years, enhancement in drug efficacy have been limited because of lack of selectivity of liposomal formulation for the cancer cell but in this study, we can find that leukemic cell receptor such as CD19, CD20, CD22 or CD38 are dual binding to a generation of bispecific antibodies for targeted delivery of PEGylated liposomal drugs to leukemic cell. Liposome targeting system follow “mix $ match” principle where bispecific antibodies were selected for specific leukemic cell receptor and Bispecific antibodies improve the targeting and cytotoxic activity/ potency of PEGylated liposomal formulation (doxorubicin) for highrisk childhood leukemia.

Doxorubicin is a PEGylated liposomal formulation used in the therapy for cancer but the issue is their efficacy which is limited and tolerability and specificity of drug to tumor cell. Finally, this study we can found that bispecific antibodies bind with leukemic cell receptor for targeted delivery of PEGylated liposomal drugs and improve the activity of doxorubicin against leukemia cell with minimal evidence of toxicity towards normal immune cell. Various AI-aligned technologies, such as computer vision, natural language processing, and pattern recognition algorithms,

Some examples of areas where AI is used include drug discovery, where it can assist with predicting outcomes of clinical trials and potential side effects of new drugs, as well as analysis of medical imagery, algorithms to spot early warning signs of disease in x-rays or MRI scans. It can also be used to analyze data collected from patient wearables or inhome sensors used in virtual hospital settings (more on that in my next trend) to provide early warning or predictive diagnosis of various conditions. Put together, all of these use cases indicate that AI and ML will continue to be a prominent trend in healthcare throughout the coming year. Nanosensors In Detecting Biomarkers Nano sensors are nanoscale devices that measure physical quantities and convert these to signals that can be detected and analyzed. Nanosensors are designed to detect a target analyte through measurable optical, electrical, magnetic, or mechanical signals. Nanosensors can also be designed to be highly sensitive. This sensitivity allows them to detect molecules at incredibly low concentrations, making them an invaluable tool in medical diagnostics.

A tiny protein binder fuses to a small hole created in the membrane of a cell—known as a nanopore—which allows ionic solution to flow through it. When the sensor recognizes a targeted molecule, the ionic flow changes. This change in flow serves as the signal from the sensor that the biomarker has been found. These new findings formulate a sensor design architecture that can be applied to a broad range of protein targets. One-day national conference was held at IIMT College of Pharmacy, Greater Noida. This conference was hosted on the topic of National Conference on Recent Advances in Pharmaceutical Sciences.
